2019 iGEM team FAFU-CHINA The Hsp70A-RbcS2 promoter enhances the expression of EGFP after heat shock initiation. It is seen that the FAFU-CHINA team in 2016 made Western blost on the expression of EGFP, at a temperature of 40 degrees Celsius. Heat shock 0, 80, 60, 40, 20min. This year (2019), we used the same promoter and EGFP for QPCR quantification, provided that the temperature was 40 ° C, heat shock 0, 40, 80, 100, 120, 140 min. The results showed that after 0, 40, 80 min, the results were the same as the 2016 FAFU-CHINA results. The new supplement condition we have this year is that the heat shock is 100, 120, 140min. The experimental results show that the relative expression of EGFP is increasing under these conditions, but the results are similar at 120 and 140 min, this may be the limit of the conditions that enhance EGFP expression. |
